Antimicrobial resistance (AMR) has become a formidable global health crisis, with the escalating threat of antibiotic resistance primarily driven by the spread of beta-lactamases. A striking example is KPC (Klebsiella pneumoniae carbapenemase), first identified in drug-resistant Klebsiella pneumoniae, which possesses the alarming ability to cleave nearly all beta-lactams, including combinational beta-lactams and beta-lactamase inhibitors, due to active-site insertions, including Ceftazidime-Avibactam. To unravel the mechanistic intricacies of these enzymes, we aim to conduct a comprehensive study using macromolecular crystallography, biophysical, biochemical, biological, and MD simulation methods.
